چکیده

Shepard (1955; 1957a,b; 1958a,b) found differences in the growth rate and pattern of mycobacterial strains in HeLa cells. Our first interest was in the use of human monocytes in measuring virulence of Mlycobacterium tuberculosis and other mycobacteria associated with pulmonary disease. As a part of this study, the trial of an established line of monocytes appeared indicated. This paper is a report of observations made on the intracellular growth of tuberele bacilli and other mycobacterial strains in the J-111 cell line of leukemic monocytes (Osgood and Brooke, 1955).

THE EFFECT OF HYPERTHERMIA ON THE DIFFERENTIATION OF LEUKEMIC CELL LINES

Treatment of human promonocytic leukemic cell line U937 with mild hyperthermia in the temperature range of 40-43°C resulted in differentiation of these cells into monocyte/macrophage-like cells in a heat dose and time dependent manner. This process was accompanied by marked morphological, functional and proliferational changes. U937 cells which normally grow in supension in the logarithmic...

Assessment of Cytotoxicity of Dimethyl Sulfoxide in Human Hematopoietic Tumor Cell Lines

Background: Dimethyl Sulfoxide (DMSO) is a solvent most broadly used as a cryopreservative agent. Antitumor effects of DMSO is a recently recognized phenomenon. In this study, cytotoxic effects of DMSO on human monocytes and T leukemic cell lines has been investigated in vitro. Methods: Human leukemic T cells (Molt-4 and Jurkat) and monocytes (U937 and THP1) were cultured in complete RPMI medi...

Multiplication of Tubercle Bacilli within Mononuclear Phagocytes in Tissue Cultures Derived from Normal Animals and Animals Vaccinated with Bcg

When monocytes derived from normal guinea pigs or rabbits were infected with tubercle bacilli and cultivated in vitro, the bacilli multiplied abundantly within the cytoplasm of these cells. By contrast, intracellular multiplication of the bacilli was retarded or completely inhibited within the monocytes of rabbits or guinea pigs vaccinated with BCG. This inhibition of growth was observed with b...

Multiplication of Mycobacterium tuberculosis Within Normal and "Immune" Mouse Macrophages Cultivated With and Without Streptomycin.

Acquired cellular immunity to infection with Mycobacterium tuberculosis is believed to reside in the capacity of mononuclear phagocytes of immunized animals to inhibit intracellular multiplication of the parasite. However, in macrophage tissue culture systems, it has been customary to employ streptomycin in the medium for the purpose of restricting extracellular, but not intracellular, growth o...

Expression Pattern of Interferon-γ in Human Leukemic T Cell Lines Following Treatment with Phytoheamagglutinin, phorbol myristate acetate and Lipopolysaccharide

Background: As a T helper type 1 (Th1) derived cytokine, Interferon gamma (IFN-γ) is an important regulator of inflammatory immune responses. Furthermore, IFN-γ plays an essential role in defense against tumors and intracellular pathogens. This study was designed to assess the pattern of IFN-γ production in human leukemic (Jurkat and Molt-4) T cell lines in vitro. Methods: Ju...
